Direct Sampling and Analysis of Environmental Swipes and Dried Blood Spot (DBS) Cards with the Advion Plate Express and Solara ICP-MS

The ability to immobilize test samples onto solid supports represents a versatile means of acquiring samples as well as a convenient means of transporting the material from the point of acquisition to the analysis/detection instrument.  In the world of nuclear safeguards and occupational/environmental health and safety, cloth swipes are a very effective means of sampling particulate matter.  By analogy, cloth and paper filters are widely used to sample particulates in gas and liquid flows.  An extremely important solid sampling matrix is the clinical use of dried blood spot (DBS) cards, employed particularly in the case of newborn babies.  Two primary scenarios exist in the sampling of the immobilized material on the fibrous matrices, the need for some form of physical or chemical means of releasing the analytes from the solid support.  In the case of ICP-MS determinations, the two most common approaches are exhaustive extraction or chemical/combustion digestion of the support.  (Laser ablation is the obvious choice in the case of single-particle analysis.)  We describe here the use of an automated, microextraction methodology for the sampling of environmental swipes as employed in nuclear safeguards applications and the direct analysis of DBS cards.  The Advion PlateExpress, originally developed for sampling of thin-layer chromatography (TLC) for organic MS determinations, has been modified to allow rapid (< 1min) sampling of these solid supports, with the eluates transported directly to the ICP-MS torch, in this case the Advion Solara.  A metal-free sampling head developed at the Oak Ridge National Laboratory allows for determinations with very low transition metal blanks; essential for the blood analysis.  We will describe the basic instrumentation and implementation of the microextraction methodology to these two important immobilization platforms.  It is believed that the general approach holds promise across a diversity of solid state immobilization sampling strategies.
